Abstract

The rs320 polymorphism of LPL gene has been reported to be associated with triglycerides, total cholesterol, HDL-C, and LDL-C. To analyze this polymorphism in Vienamese populations, the study aimed to optimize the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method with primers, thermal cycles, and restriction enzyme
which were selected and tested to identify opimal condition of PCR-RFLP method. The GG, TG, and TT genotypes by PCR - RFLP methods were confirmed by the sequencing method. The
experimental results determined the annealing temperature of 57 oC for PCR, the minimum 3 units of HindIII for incubation with PCR product. In conclusion, this optimal PCR-RFLP
method can be applied to genotype LPLrs320 in large populations to investigate the association between the rs320 polymorphism and dyslipidemia.
