SIMULTANEOUS DETERMINATION OF TYROSINE AND TRYPTOPHAN BY FLUORESCENCE KINETICS OF RUTHENIUM(II) POLYPYRIDYL COMPLEX

Abstract

   A fluorescence-kinetic method has been developed and validated for simultaneousdertemination of tyrosine (Tyr) and tryptophan (Trp) in protein based on theirphotoredox reactions with rutheni(II) polypyridine complexes. The proposed methodis simple and accurate with a recovery of 99 - 104 %. The detection limit is 2.91  10-5 M with a RSD < 3 % (n=6). The results of Tyr and Trp analyses in protein samplesusing the proposed method and HPLC are identical. Therefore, the fluorescencekineticmethod is applicable to determine Tyr and Trp.
