The risk of exposure to aminoglycoside antibiotic resistance genes increases the incidence of infections caused by drug-resistant bacteria, leading to clinical treatment failure with kanamycin. This situation necessitates the urgent implementation of new strategies to control antibiotic resistance. In this study, we developed a highly sensitive fluorescence “on-off” nanosensor for kanamycin detection based on the interaction between g-C₃N₄ and gold nanoparticles (AuNPs). The addition of AuNPs to the g-C₃N₄ solution resulted in fluorescence quenching (off effect). However, in the presence of kanamycin, the fluorescence signal was restored due to the analyte binding to AuNPs and releasing g-C₃N₄ (on effect). Under optimal conditions (6 mM Na₂SO₄, pH 7.0, incubation time of 15 minutes), the method demonstrated a wide linear range from 10 nM to 500 nM with high accuracy (RSD < 5%). This approach was successfully applied for kanamycin detection in pharmaceutical samples, highlighting its potential for antibiotic residue monitoring and antibiotic resistance mitigation.
