In vitro shoot regeneration of Adenosma indiana (Lour.) Merr.

Abstract

This paper presents the obtained results of in vitro shoot regeneration of Adenosma indiana (Lour.) Merr. Shoot tips and stem of native Adenosma indiana were sterilized with HgCl2 0.1% in 5-10 minutes. Results indicated that highly effective sterilization occurred with HgCl2 in 5 to 10 minutes for reducing the contamination rate and increasing the survival rate to 60.35% for stem explants and 57.44% for shoot tips. After 4 weeks of culture, the best regeneration buds achieved on MS medium supplemented with 0.5 mg/l BAP (46.70% of stem explant regenerating shoots and 1.8 shoots/stem explant). The shoot multiplication from in vitro shoots achieved quickly on MS medium supplemented alone with BAP and kinetin or in combination with 0.5 mg/l BAP with NAA or IBA. After 6 weeks of culture, the multiplication coefficent was greatest on MS medium supplemented in combination 0.5 mg/l BAP with 0.3 mg/l IBA. The average number of shoots per explant was 10.03.
